Summary

Altered metabolism is a hallmark of many disease states, including cardiovascular and musculoskeletal conditions. Importantly, metabolic profiles may be patient-specific and may for example indicate susceptibility to treatment or malignancy. Yet, efficient methods to characterize the metabolic state of tissue and organs noon-invasively are lacking.

Full description

While Magnetic Resonance Imaging is well established to assess anatomy, structure and function in patients and in animal models of human disease alike, (routine) MR Spectroscopy tools to investigate metabolic alterations are far less common. Long scan times and low spatial resolution associated with MRS experiments limit the wider applicability of this versatile technique. SPectroscopic Imaging by exploiting spatiospectral CorrElation (SPICE) is a novel and emerging technique that promises to overcome some of the limitations and allow for high-resolution MR spectroscopic imaging (MRSI) at reasonable scan time. Here, the signals are represented by a low-rank model capturing the spatiospectral correlation of the data. This model implies that high-dimensional MRSI data reside in a very low dimensional subspace. Special data acquisition schemes are then used in combination with dedicated reconstruction techniques to recover high resolution MRSI images from undersampled k-space data. SPICE, although promising, has so far mainly been used in proof-of-concept studies including in brain (1H-MRSI) and skeletal muscle (31P-MRSI). We therefore propose to develop SPICE as a tool for advanced metabolic profiling of skeletal muscle and the heart.

Under the direct supervision of Profs Schneider (Physics) and Plein (Clinical) and as part of a larger translational research group, the student will work to deliver the following specific objectives:

develop the SPICE framework;

implement bespoke metabolic imaging sequences on the clinical MR system in the university’s Advanced Imaging Centre;

validate the technique against conventional MRSI methods;

apply clinical proof-of-concept studies in patients with musculoskeletal, metabolic and cardiovascular disease.

To support the application process or if you want to know more about our non-clinical PhDs a recording of our 2023 webinar can be found via the links below

**The deadline dates in the webinar are out of date but are correct in the stand-alone slide deck**

NIHR Leeds BRC Non Clinical PhD Scheme webinar - YouTube

with a copy of the slide deck used at

Leeds BRC Non Clinical PhD Webinar FINAL updated slide deck March 24 - YouTube
